| Fig. 1 The protein kinase A pathway: 1) Ligand binds to G-protein coupled receptor (GPCR) on the plasma membrane. This catalyzes exchange ofGTP for GDP on the α subunit of the associated heterotrimeric G protein, with dissociation of the α subunit from the βγ dimer; 2) The α subunit binds to adenylyl cyclase (AC) and activates its enzymatic activity. Activated AC produces cyclic AMP (cAMP) from ATP; 3) cAMP binds to the regulatory subunits (R) of protein kinase A (PKA) which allows for release of the catalytic subunits (c) of PKA, that then mediate serine-threonine phosphorylation of target molecules, including the transcription factor CREB (cAMP response element-binding protein).

RMCs were pre-incubated with 100 nmol/L rapamycin ( + RA) for 2 h before stimulating with high glucose (HG, 30 mmol/L glucose), and incubated for 2 h for detection of Egr1 and 24 h for detection of TLR4. Protein expression was determined by western blot. * p < 0.05, * * p < 0.001 vs. RMCs cultured with high glucose media. #p < 0.001 vs. RMCs cultured with high glucose media. All results represent means ± SD obtained from 3 independent experiments and normalized to GAPDH. TLR4: toll like receptor 4; Egr1, early growth response factor 1; GAPDH, glyceraldehyde-3-phosphate dehydrogenase.
